Background
Piccolo (also known as Aczonin) is a large, multidomain scaffolding protein localized at the presynaptic active zone. It plays a central role in organizing the cytomatrix at active zones (CAZ) and coordinating synaptic vesicle trafficking, exocytosis, and endocytosis. Piccolo interacts with proteins involved in actin dynamics and clathrin-mediated endocytosis, including Abp1 and dynamin, thereby regulating synaptic vesicle recycling. It is often co-expressed with Bassoon, another CAZ protein, to maintain synaptic integrity and plasticity. Disruption of Piccolo expression or function has been linked to neurodevelopmental and neurodegenerative disorders, including schizophrenia and Parkinson’s disease. Its role in synaptic maintenance and plasticity makes Piccolo a promising target for understanding and treating synaptic dysfunction in neurological diseases.
