  • Background:

    JC-1 is a fluorescent dye that is able to pass across the mitochondrial membrane. It is used to measure mitochondrial membrane potential. At low membrane potentials, it remains a monomer and emits green-fluorescent light (λex=485 nm, λem=520 nm). At higher membrane potentials, it forms "J-aggregates", which emit red-fluorescent light (λex=520-570 nm, λem=570-610 nm). The ratio of red/green fluorescence is based only on the membrane potential, and not on mitochondrial size, shape or density.
