Background:
Autophagy-related protein 2A (ATG2A) is a core component of the autophagy machinery and a member of the ATG2 protein family, essential for maintaining cellular homeostasis under stress conditions. ATG2A plays a pivotal role in autophagosome biogenesis by facilitating membrane expansion and lipid transfer at the phagophore assembly site. In parallel, ATG2A regulates lipid droplet morphology and dispersion, linking autophagy to lipid metabolism—two processes increasingly recognized as central to neuronal health and disease. In the context of neurodegenerative disorders, ATG2A is emerging as a key player in the cellular response to proteotoxic stress. Impaired autophagy and disrupted lipid homeostasis are hallmark features of diseases such as Alzheimer’s, Parkinson’s, and Huntington’s, where accumulation of misfolded proteins and dysfunctional organelles contributes to neuronal death. By coordinating membrane dynamics and lipid trafficking, ATG2A supports the clearance of toxic aggregates and sustains energy balance in neurons.
