  • Background:

    UVRAG (Ultraviolet irradiation resistance-associated gene) encodes a multifunctional protein that plays a pivotal role in cellular homeostasis, particularly through its regulation of autophagy and endosomal trafficking. As a key activator of the Beclin1–PI3KC3 complex, UVRAG promotes autophagosome formation and maturation, facilitating the clearance of damaged organelles and misfolded proteins—processes that are often disrupted in neurodegenerative diseases. In addition to its autophagic functions, UVRAG contributes to genomic stability by participating in DNA repair and maintaining centrosome integrity. These roles are especially critical in neurons, which are highly sensitive to oxidative stress, DNA damage, and impaired proteostasis. Dysregulation of UVRAG has been linked to pathological features common in neurodegenerative disorders such as Alzheimer’s disease, Parkinson’s disease, and amyotrophic lateral sclerosis. Impaired UVRAG function can lead to defective autophagy, accumulation of toxic protein aggregates, and increased neuronal vulnerability.
