  STAT3-IN-20 (Compound 40), with an IC50 of 0.65 ?M, is a selective inhibitor targeting the SH2 domain of STAT3, thereby impeding its phosphorylation, nuclear translocation, and subsequent gene transcription. The compound demonstrates antiproliferative effects on DU145 and MDA-MB-231 cancer cell lines, which have overactivated STAT3, with IC50 values of 2.97 ?M and 3.26 ?M, respectively. Moreover, STAT3-IN-20 induces cell cycle arrest and apoptosis [1].
