Background:
Phosphodiesterases (PDEs) play an important role in the dynamic regulation of the second messengers cAMP (cyclic adenosine monophosphate) and cGMP (cyclic guanosine monophosphate) signaling, by hydrolyzing them. The PDE superfamily is composed of 11 families, with PDE4,7 and 8 being cAMP-specific hydrolases, and thus regulating positive and negative responses to cAMP. PDE3B, like PDE3A, has a high affinity for both cAMP and cGMP, but differs in tissue expression pattern. In addition to vascular smooth muscles and kidney, it can also be found in adipocytes, hepatocytes, β cells, and T cells and macrophages. In cardiomyocytes it is in the T tubules, linking it to a metabolism regulation role. For instance, in adipocytes, insulin increases membrane-bound phosphorylated PDE3B, which interacts with IRS-1 (insulin receptor substrate 1) and other proteins. Inhibitors for PDE3A and PDE3B showed some promise for cardiovascular disease treatment but can result in side effects such as arrhythmia. The development of PDE3B-specific inhibitors may prove crucial in the treatment of ischemia and reperfusion injury.Description:
