  • Background:

    Rab5 is a 24 kDa GTPase that regulates early endosome dynamics and endocytic trafficking. It is ubiquitously expressed and localizes to early endosomes and the plasma membrane. Rab5 controls the fusion of endocytic vesicles with early endosomes and mediates homotypic endosome fusion. It recruits effectors such as EEA1 and Rabaptin-5 to facilitate vesicle docking and fusion. In the nervous system, Rab5 is essential for receptor internalization, synaptic vesicle recycling, and neurotrophin signaling. Dysregulation of Rab5 activity can lead to endosomal dysfunction, impaired signaling, and neurodegeneration. It is widely used as a marker of early endosomes and is a key player in maintaining neuronal homeostasis.
