Background
Rab4 is a 25 kDa GTPase of the Ras superfamily that regulates early endosomal recycling. It cycles between an inactive GDP-bound state and an active GTP-bound state, recruiting effector proteins to intracellular membranes. Rab4 modulates receptor recycling, including transferrin and glucose transporter (GLUT4) trafficking. Overexpression of Rab4 alters receptor distribution, leading to accumulation in post-endosomal compartments. Its activity is regulated by phosphorylation and post-translational lipid modifications, which influence membrane association and subcellular localization. In neurons, Rab4 is involved in synaptic vesicle recycling and receptor trafficking. Dysregulation of Rab4 may impair endosomal sorting and contribute to synaptic dysfunction in neurodegenerative diseases such as Alzheimer’s and Parkinson’s.
