Test principle
This EIA kit uses the Sandwich-EIA principle. The micro EIA plate provided in this kit has been pre-coated with an antibody specific to Human BACE1. Standards or samples are added to the micro EIA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human BACE1 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human BACE1, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human BACE1. You can calculate the concentration of Human BACE1 in the samples by comparing the OD of the samples to the standard curve.

Description
This ELISA test kit or EIA Enzyme immuno assay is an enzyme linked immuno assay supplied as coated 96 well plates with antigen or antibody and needs to be stored at +4°C. Enzymes are cleaving the substrate. If the substrate is DNA they are called restriction enzymes. Activating enzymes will cut off the domain that is biological active to become functional.

Properties
MeSH E05 478 566 350 170 Enzyme-Linked Immunosorbent Assay, Human proteins, cDNA and human recombinants are used in human reactive ELISA kits and to produce anti-human mono and polyclonal antibodies. Modern humans (Homo sapiens, primarily ssp. Homo sapiens sapiens). Depending on the epitopes used human ELISA kits can be cross reactive to many other species. Mainly analyzed are human serum, plasma, urine, saliva, human cell culture supernatants and biological samples.

Scope note:
Immunologic techniques based on the use of: (1) enzyme-antibody conjugates; (2) enzyme-antigen conjugates; (3) antienzyme antibody followed by its homologous enzyme; or (4) enzyme-antienzyme complexes. These are used histologically for visualizing or labeling tissue specimens.
