  • Background:

    The low affinity immunoglobulin E (IgE) Fc receptor, also known as CD23, is a 45kD type II transmembrane glycoprotein. CD23 contains a C-type lectin domain in the extracellular C-terminal region and belongs to group II of the type II lectins. CD23 is a receptor for IgE but can also bind to other ligands such as CD21, CD11b and CD11c. CD23 primarily functions as a B-cell growth and activation factor. It also regulates the synthesis of IgE through binding of IgE and CD21. Two isoforms of CD23 have been described, CD23a and CD23b, which are expressed on different cell types. CD23a expression is restricted to resting B-cells, whereas CD23b is weakly expressed on activated B-cells and other haematopoietic cells. CD23 is also expressed on B-cell chronic lymphocytic leukemia (CLL) cells and is useful as a marker to differentiate CLL from mantle cell lymphoma by flow cytometry. A soluble form of CD23 (sCD23) also exists which is produced by proteolytic cleavage of the membrane bound form. Studies suggest that levels of serum sCD23, in cases of B-CLL, correlate with tumour activity and are an important indicator for early treatment.

    5E3 is a mouse monoclonal IgG1 antibody derived by fusion of SP2/0 mouse myeloma cells with splenocytes from a BALB/c mouse immunized with Daudi cells, a Human Burkitt’s lymphoma cell line.

  • Assay Principle:

    The 5E3 antibody is suitable for the detection of Human CD23 antigen by flow cytometry. Human tonsil is suitable as a positive control. Optimal antibody dilutions for the different applications should be determined by titration.

  • Storage Conditions:

    The antibody is shipped at ambient temperature and may be stored at +4°C; For prolonged storage prepare appropriate aliquots and store at or below -20°C; Prior to use, an aliquot is thawed slowly in the dark at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2); Repeated thawing and freezing should be avoided; Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day; If a slight precipitation occurs upon storage, this should be removed by centrifugation; It will not affect the performance or the concentration of the product
