Human HCC-1 (CCL14) Recombinant

  • Background:

Hemofiltrate C-C chemokine (HCC) -1 is a recently described monocyte chemoattractant. CCL14 (also known as HCC-1) belongs to the CC chemokine family. Its mature propeptide is a low- affinity agonist of CCR1 that is converted to a high- affinity agonist of CCR1 and CCR5 on proteolytic processing by serine proteases. Determination of the amino acid sequence of HCC-1 revealed four cysteine residues in positions characteristic of the C-C chemokine family, and comparison with the sequences of other chemokines revealed that HCC-1 was most homologous to MIP-1a. However, several functional properties of HCC-1 were atypical of chemokines. Unlike other chemokines, HCC-1 was expressed constitutively in a number of tissues and was present at high concentrations in normal human plasma. In addition, HCC-1 was reported not to be chemotactic for leukocytes.
  • Description:

Recombinant HCC-1 is a disulfide-linked monomeric protein consisting of 73 amino acid residues and migrates as an approximately 9 kDa protein under non-reducing conditions and reducing conditions in SDS-PAGE. Optimized DNA sequence encoding human HCC-1 (CCL14) mature chain was expressed inE. coli.
  • UniProt:

    Q16627
  • Endotoxin:

    < 0.1 ng/µg (1 EU/µg), using the LAL gel clot method.
  • Purity:

    ≥ 98% by SDS-PAGE and HPLC
  • Format:

    Lyophilized protein
  • Reconstitution:

Reconstitute at 0.1-1.0 mg/ml in distilled water. This solution can then be diluted into other buffers. To maximize product collection from vial surface, vortex briefly and then spin down to recollect the liquid.
  • Shipping Conditions:

    Gel Packs or Dry Ice
  • Storage Conditions:

The lyophilized protein is stable for at least 2 years from date of receipt when stored at -20°C. Upon reconstitution, store in working aliquots at +4°C for up to one month, or at -20°C for up to six months, in the presence of a carrier protein. Avoid repeated freeze/thaw cycles.
  • Calculated Molecular Weight:

    9 kDa
  • Formulation:

Lyophilized from 0.2 µm filtered solution of 40 mM NaCl, 10 mM phosphate buffer, pH 7.0.
