SENP7, a versatile protease, positively regulates the cGAS-STING pathway by desumoylating CGAS, enhancing its DNA-binding and activation. Specific to SUMO2 and SUMO3, SENP7 proficiently deconjugates poly-SUMO2 and poly-SUMO3 chains, dynamically regulating protein sumoylation. While less efficient in processing full-length SUMO proteins, SENP7's role in modulating the cGAS-STING pathway highlights its importance in innate immune responses.
