Mouse anti Desmin
- Background: Desmin is a 53 kDa intermediate filament protein that exhibits a high degree of tissue specificity, its expression being predominantly confined to all types of muscle cells (cardiac, skeletal and smooth muscle). Regulation of desmin expression is stage and tissue-specific, since it is induced during terminal development. In skeletal en cardiac muscle cells desmin is localized in the Z-disk region and at the intercalated disk. The expression pattern of desmin in smooth muscle is much more heterogenous. Coexpression of vimentin and desmin has been observed in tumors derived from muscle tissue, i.e. rhabdomyosarcomas and leiomyosarcomas. Furthermore, during myocard dysfunction dramatic changes in the distribution of desmin have been observed.
- CAS Number: 9007-83-4
- UniProt: P17661
- Host: Mouse
- Species Reactivity: Human, Hamster, Rat, Chicken
- Isotype: IgG1
- Clone: D33
- Type: Primary Antibodies
- Source: D33 is a mouse monoclonal IgG1 antibody derived by fusion of mouse myeloma cells with spleen cells from a BALB/c mouse immunized with purified desmin from a leiomyosarcoma.
- Applications: ICC, IHC (frozen), IHC (paraffin), WB
- Field of Research: Cell Biology
- Assay Principle: The D33 antibody is suitable for the detection of desmin by Western blotting and detects a band of 53kDa in muscle tissue extracts. The antibody is also suitable for the detection of desmin by immunocytochemistry on permeabilized cells, and Immunohistochemistry on frozen and formalin fixed, paraffin embedded tissues. The antibody works on paraffin-embedded material without pretreament with proteolytic enzymes, but the section should be pre-treated using heat mediated antigen retrieval with Tris-buffer containing 1 mM EDTA, pH 9,0 for 20 mins. Optimal antibody dilutions for the different applications should be determined by titration.
- Form: Each vial contains 100 µL 1mg/mL purified monoclonal antibody in phosphate buffered saline (PBS) containing 0,09% sodium azide
- Precautions: This product is intended FOR RESEARCH USE ONLY, and FOR TESTS IN VITRO, not for use in diagnostic or therapeutic procedures involving Humans or animals. This product contains sodium azide. To prevent formation of toxic vapors, do not mix with strong acidic solutions. To prevent formation of potentially explosive metallic azides in metal plumbing, always wash into drain with copious quantities of water. This datasheet is as accurate as reasonably achievable, but Nordic-MUbio accepts no liability for any inaccuracies or omissions in this information.

- Storage Conditions: The antibody is shipped at ambient temperature and may be stored at +4°C; For prolonged storage prepare appropriate aliquots and store at or below -20°C; Prior to use, an aliquot is thawed slowly in the dark at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2); Repeated thawing and freezing should be avoided; Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day; If a slight precipitation occurs upon storage, this should be removed by centrifugation; It will not affect the performance or the concentration of the product
