Heme-oxygenase is a ubiquitous enzyme that catalyzes the initial and rate-limiting steps in heme catabolism yielding equimolar amounts of biliverdin, iron and carbon monoxide. Biliverdin is subsequently converted to bilirubin and the free iron is sequestered to ferritin (1). These products have important physiological effects as carbon monoxide is a potent vasodilator; biliverdin and bilirubin are potent antioxidants; and the free iron increases oxidative stress and regulates the expression of many mRNAs (2). There are three isoforms of heme-oxygenase, HO-1, HO-2 and HO-3; however HO-1 and HO-2 are the major isoforms as they both have been identified in mammals (3). HO-1, also known as heat shock protein 32, is an inducible isoform activated by most oxidative stress inducers, cytokines, inflammatory agents and heat shock. HO-2 is a constitutive isoform which is expressed under homeostatic conditions. HO-1 is also considered to be a cytoprotective factor in that free heme is highly reactive and cytotoxic, and secondly, carbon monoxide is a mediator inhibiting the inflammatory process and bilirubin is a scavenger for reactive oxygen, both of which are the end products of heme catalyzation (4). It has also been shown that HO-1 deficiency may cause reduced stress defense, a pro-inflammatory tendency (5), susceptibility to atherosclerotic lesion formation (6), endothelial cell injury, and growth retardation (7). Up-regulation of HO-1 is therefore said to be one of the major defense mechanisms of oxidative stress (4).

About
Polyclonals can be used for Western blot, immunohistochemistry on frozen slices or parrafin fixed tissues. The advantage is that there are more epitopes available in a polyclonal antiserum to detect the proteins than in monoclonal sera. Rabbits are used for polyclonal antibody production by StressMark antibodies. Rabbit antibodies are very stable and can be stored for several days at room temperature. StressMark antibodies adds sodium azide and glycerol to enhance the stability of the rabbit polyclonal antibodies. Anti-human, anti mouse antibodies to highly immunogenic selected peptide sequences are" monoclonal like" since the epitope to which they are directed is less than 35 amino acids long. Rats are used to make rat monoclonal anti mouse antibodies. There are less rat- than mouse clones however. Rats genes from rodents of the genus Rattus norvegicus are often studied in vivo as a model of human genes in Sprague-Dawley or Wistar rats.

Polyclonal, Rabbit, anti-, Percp, anti, antibody to, antibodies, Peridinin chlorophyll protein (PerCP) has a 677 nanometer maximum emission, red, when excited at 488 nanometers and the Rabbit Anti- HO-2 Polyclonal: PerCP is detected on the FL3 detector of a FACSCalibur or FACScan. A PerCP dye is also available (PerCP-Cy5.5, also written Cy5.5-PerCP). PerCP is not suited for the high-powered lasing (>150mW) applications, such as on a MoFlo, due to it photobleaching (features/ qualities/ traits). PerCP conjugates can only be received/be gotten from the Becton Dickinson/Pharmingen group of companies or from StressMark antibodies. Polyclonal antibodies (pAbs) are mostly rabbit or goat antibodies that are secreted by different B cells, whereas monoclonal antibodies come from a single N cell lineage. Pabs are a collection of immunoglobulin molecules that react against a specific antigen, each identifying a different epitope.
