Background
PRMT9 (protein arginine methyltransferase 9) is a type II methyltransferase, as it generates SDMA (symmetrically di-methylated arginine) . Arginine methylation of proteins is a common post-translational modification, particularly in RNA-binding proteins. PRMT9 contains two S-adenosylmethionine binding motifs and three TPRs (tetratricopeptide repeats) . It is a modulator of the splicing factor SAP145/SAP149 complex. PRTM9 has increased activity in blast and leukemia stem cells (LSCs) derived from patients with AML (acute myeloid leukemia), indicating a potential role in cancer cell survival and proliferation. Indeed, PRTM9 inhibition increased DNA damage in these cells, and type I INF (interferon) response. The use of combinatory therapy of a PD-1 (programmed cell death protein 1) /PD-L1 (programmed death-ligan 1) inhibitor with a PRTM9 inhibitor may prove beneficial in oncology. Additionally, the PRMT9/SAP415 axis was also implicated in autosomal recessive intellectual disability (ARID), bringing into light a new role of this protein.
Description
The PRMT9 Homogenous Assay Kit is designed to measure the activity of PRMT9 (protein arginine methyltransferase 9) for screening and profiling purposes. The PRMT9 Homogeneous Assay Kit comes in a convenient format 384-well AlphaLISA® format, with enough purified recombinant PRMT9, S-adenosylmethionine, primary antibody, SAP145 (amino acids 401-550) and buffers for 400 enzyme reactions.Figure1: Illustration of the assay principle.First, the PRMT9 enzyme is pre-incubated with the inhibitor, and the substrate mixture is added to the wells to initiate the methyltransferase reaction. Next acceptor beads, a highly specific primary antibody directed against methylated peptides, and donor beads are added, and Alpha-counts are measured. Contraindications
The PRMT9 Homogenous Assay Kit is compatible with up to 1% final DMSO concentration.Avoid green and blue dyes that absorb light in the AlphaScreen signal emission range (λ=520-620 nm), such as Trypan Blue.Avoid the use of potent singlet oxygen quenchers such as sodium azide (NaN3) or metal ions (Fe2+, Fe3+, Cu2+, Zn2+and Ni2+) .The presence of > 1% RPMI 1640 culture medium leads to a signal reduction due to the presence of excess biotin and iron in this medium. Media like MEM, which lacks these components, does not affect AlphaScreen assays.
