  PF-04449913 is a potent and orally bioavailable smoothened (SMO) inhibitor. PF-04449913 binds to human SMO with an IC₅₀ of 4 nM. It has been shown to attenuate leukemia stem cell self-renewal and cell cycle progression in primary acute myeloid leukemia cells and in an in vivo Drosophila model.
