  • Assay Protocol:

    1. After the peroxidase reaction, wash membrane thoroughly with PBS or Tris-HCl saline containing 0.1 % Tween 20. DO NOT ADD THIMEROSAL, azide or phenolic compounds as preservative in any buffers. 2. Cover membrane with TMBM solution and incubate with gentle rocking for 5-30 minutes till aqua blue color bands appear. (If bands appear immediately; the concentration of peroxidase labeled probe is too high and should be further diluted). 3. Stop reaction by washing thoroughly in distilled or deionized water. DO NOT WASH WITH BUFFER OR TAP WATER. DO NOT INTRODUCE ANY ACID AT THIS STAGE, THE BANDS WILL TURN YELLOW.
