  • Background
BioVision’s human NRG-1 ELISA Kit is based on the standard principle of a sandwich enzyme-linked immunosorbent assay. A mouse monoclonal antibody specific for human NRG-1 is coated on a 96-well plate. Standards and test samples are added to the wells and NRG-1 present in a sample is bound by the immobilized antibody. A biotinylated polyclonal detection antibody from goat specific for NRG-1 is added subsequently. After washing away the unbound biotinylated antibody with PBS or TBS buffer, HRP conjugate is added to the wells. The wells are again washed with PBS or TBS buffer to remove the unbound conjugates. HRP substrate TMB is used to visualize the HRP enzymatic reaction. TMB is used by HRP to produce a blue product that changes into yellow after adding acidic stop solution. The density of yellow color is proportional to the human NRG-1 captured onto the plate. This ELISA kit shows no detectable cross-reactivity with related proteins. Detection Range: 62.5 pg/ml – 4000 pg/ml. Sensitivity: < 10 pg/ml. Coefficient of variation: 5% (Intra-assay) and 7% (Inter-assay).

Modern humans (Homo sapiens, primarily ssp. Homo sapiens sapiens). Depending on the epitopes used human ELISA kits can be cross reactive to many other species. Mainly analyzed are human serum, plasma, urine, saliva, human cell culture supernatants and biological samples.

ELISA test kits can be sandwich ELISA. In sandwich ELISAS Enzyme-linked immunosorbent assay 9we use captor and detector antibodies. In single antibody ELISAs the antigen is coated and only a detector antibody is used. Traditional competition antigen ELISAs are coated with a captor antibody and a competitive antigen is labelled with the chromogen. In this case the ratio on your graph will be inverse proportional with the antigen present in your sample. A sign detected by the Colorimetric detection OD optical density absorbance at 450 nm between 0 and 4 with an ELISA reader for 450 nm absortion. Scope note: An immunoassay utilizing an antibody labeled with an enzyme marker such as horseradish peroxidase. While either the enzyme or the antibody is bound to an immunosorbent substrate, they both retain their biologic activity; the change in enzyme activity as a result of the enzyme-antibody-antigen reaction is proportional to the concentration of the antigen and can be measured spectrophotometrically or with the naked eye. Many variations of the method have been developed.
