Scientific contextMAP3K8 is a protein serine/threonine kinase that can activate both MAPK and JNKK pathways. It is required for TLR4 activation of the MEK/ERK pathway. It can activate NF-kB1 by stimulating the proteolysis of its inhibitor, p105, and promote the production of TNF-alpha and IL-2 during T-cell activation. It has been shown to mediate Raf-inhibiting drug resistance in BRAF(V600E) cell lines. MAP3K8 may also play a role in the cell cycle.
