Difamilast

Atopic dermatitis is a chronic and fluctuating pruritic inflammatory skin condition characterized by pruritic eczema and is also marked by exacerbations and remissions. This skin disease occurs most frequently in children and can have an onset as early as within the first 2 years of life. Difamilast is a selective PDE4 inhibitor and the inhibition of phosphodiesterase-4 (PDE4) reduces proinflammatory mediators and cytokines in atopic dermatitis. As difamilast's mechanism of action involves the inhibition of PDE4, the inhibition leads to an increase in intracellular cyclic AMP and a subsequent decrease in the production of cytokines and chemokines. The drug is indicated for the topical treatment of mild to moderate atopic dermatitis in adults and pediatric patients 2 years of age and older. It is approved in Japan and the United States.
