Acetate

The Acetate Assay is a 100-well quantitative colorimetric assay for acetate measurement. It can be used with serum, plasma, cell lysates and tissue extracts and is suitable for samples from different species. Detection is performed using a microplate reader at OD 450 nm. The assay has a reported sensitivity of 20 µM, with acetate quantification below 1 nmole per well under the manufacturer's stated conditions. The kit is stored at −20°C, shipped using a gel pack, and has a stated shelf life of one year from the date of delivery.

How the Acetate Assay Works

The assay uses an enzyme-coupled reaction sequence to convert acetate into a measurable colorimetric signal.

Acetate is first phosphorylated, converting ATP to ADP. The generated ADP is then used in a coupled reaction involving glucose and glucose-6-phosphate formation. This reaction ultimately converts NAD to NADH. NADH then participates in the reduction of a nearly colorless tetrazolium compound to a strongly colored formazan product. The resulting absorbance is measured at 450 nm.

This enzymatic coupling provides a quantitative relationship between the measured color intensity and acetate concentration.
