Description
ViralBoost Reagent is a small-molecule cocktail designed to enhance high-titer retroviral and lentiviral production. By regulating viral RNA packaging at the transcriptional level, it can increase viral particle production by up to 10-fold. Its easy-to-use protocol makes it suitable for virus packaging across a range of production scales.
Applications
ViralBoost Reagent is designed to enhance virus packaging and high-titer viral production, supporting efficient production of retroviral and lentiviral particles. It can increase viral particle production by up to 10-fold, making it suitable for applications requiring enhanced retrovirus or lentivirus production.
Assay Protocol
The ViralBoost Reagent (500X) protocol is performed over approximately 5 days, starting with the preparation and plating of HEK 293T cells, followed by transfection with the viral vector and packaging mix using NanoFect. The following day, the culture medium is replaced with fresh medium supplemented with ViralBoost Reagent to enhance viral particle production. Viral supernatant is then collected, clarified by centrifugation, filtered, and concentrated using a retrovirus/lentivirus precipitation solution. The resulting viral particles are recovered, resuspended in cold PBS or DMEM, aliquoted, and stored at −80°C until use. ViralBoost can be removed during subsequent viral concentration or purification procedures.
