Description
This antibody recognizes the D1 domain of an N-glycosylated glycoprotein of 120kDa with intra-chain disulfide bonds, identified as CD50 or ICAM3 (Leucocyte Workshop VI) . It is the major ligand for LFA-1 (CD11a/CD18) and may have signaling role to increase adhesion. It is expressed on thymocytes and T lymphocytes and is resistant to treatment with phosphatidylinositol (PI) phospholipase C. This CD50 antibody is excellent for staining of formalin/paraffin tissues.
Reactivity
Human
Immunogen
Stimulated human leukocytes were used as the immunogen for this CD50 antibody.
Clonality
Monoclonal
Clone
186-2G9
Conjugation
Unconjugated
Field of Research
Immunology & Inflammation
Purification
Protein G affinity chromatography
Dilution
Blocks binding of CD11a (LFA-1) to CD50 (ICAM-3) (order BSA/sodium azide-free format), Immunofluorescence: 0.5-1ug/ml, Immunohistochemistry (FFPE) : 0.5-1ug/ml for 30 min at RT
Storage Conditions
Maintain refrigerated at 2-8°C for up to 2 weeks. For long term storage store at -20°C in small aliquots to prevent freeze-thaw cycles.
Notes
For research use only.
Applications Notes
The concentration stated for each application is a general starting point. Variations in protocols, secondaries and substrates may require the CD50 antibody to be titered up or down for optimal performance.1. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes.2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.
