  • Background:

    N-terminal arginylation is a conserved post-translational modification that adds arginine residues to proteins, primarily via the action of arginyltransferase ATE1. This modification plays a pivotal role in embryonic development, cytoskeletal regulation, and cellular stress responses. In the context of neurodegeneration, N-terminal arginylation is essential for maintaining neuronal proteostasis and responding to endoplasmic reticulum and oxidative stress. It facilitates the degradation of misfolded proteins and modulates the actin cytoskeleton, both of which are critical for synaptic function and neuronal survival. Disruption of arginylation pathways has been linked to neurodevelopmental disorders and may contribute to the progression of diseases such as ALS and Alzheimer’s.
