  • Background:

    N-terminal arginylation is a post-translational modification catalyzed by arginyltransferase ATE1, in which an arginine residue is added to the N-terminus of proteins. This process plays a critical role in protein quality control, cellular stress responses, and neurodevelopment. In the nervous system, N-terminal arginylation regulates the degradation of misfolded or damaged proteins via the N-degron pathway, thereby maintaining proteostasis—a function essential for neuronal survival. Recent studies have linked impaired arginylation to neurodegenerative diseases such as Alzheimer’s and Parkinson’s, where accumulation of misfolded proteins and disrupted stress responses are key pathological features.
