Description
UreaGel Concentrate is one of three components of the UreaGel System, together with UreaGel Diluent and UreaGel Buffer. The system provides a convenient and dependable way to prepare gels of varying percentages, including commonly used formulations up to 20% monomer (19:1 acrylamide/bisacrylamide).
Assay Protocol
Determine the required volumes of UreaGel Concentrate, UreaGel Buffer, and UreaGel Diluent according to the desired gel percentage and total casting volume. Calculate the volumes using Vc = (Vt × X) / 25, Vb = 0.1 × Vt, and Vd = Vt − (Vc + Vb). Combine the calculated components in an Erlenmeyer flask and swirl gently. Add 40 µL of TEMED per 100 mL of gel casting solution, followed by 0.8 mL of freshly prepared 10% ammonium persulfate per 100 mL. Swirl gently, cast the gel, insert the comb, and allow the gel to polymerize for 1–2 hours.
