  • Background:

    The reactivity of the antiserum is restricted to C-reactive protein. In immunoelectrophoresis and radial immunodiffusion (Ouchterlony), using various antiserum concentrations against fresh normal Human plasma a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified C-reactive protein. No reaction is obtained with any other plasma protein component or serum. In precipitating techniques as immunoelectrophoresis and single or double radial immunodiffusion to identify the presence of C-reactive protein in Human plasma or other body fluids or to determine its concentration. The presence of non precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in more sensitive non-precipitating antibody-binding techniques if proper controls are included.

    C-reactive protein is an acute phase protein, synthesized in the liver. Its molecular weight is about 140,000. It occurs in serum of healthy men in low concentrations, <1.2 μg/mL. In cases of bacterial infection, surgical trauma and Hodgkins disease, he concentration is highly increased. It may reach values of 0.5 mg/mL. Freund’s complete adjuvant is used in the first step of the immunization procedure.
  • Assay Principle:

    Precipitation assays. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifµgation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4°C, not refrozen, and preferably used the same day.
