Relacorilant

Relacorilant is a first-in-class, selective glucocorticoid receptor antagonist. Originally investigated to manage the metabolic and endocrinologic complications of hypercortisolism, such as Cushing’s syndrome, its clinical utility has pivoted toward oncology as a potent sensitizing agent. By competitively blocking the glucocorticoid receptor, relacorilant prevents cortisol from activating anti-apoptotic survival pathways, thereby restoring the efficacy of cytotoxic therapies in the context of resistant cancers. Relacorilant was approved by the US FDA on March 25, 2026, for use in combination with [paclitaxel] in patients with certain platinum-resistant cancers.
