MRAP protein crucially modulates melanocortin receptors (MC1R, MC2R, MC3R, MC4R, and MC5R), enhancing ligand sensitivity and amplifying cAMP generation. Its role extends to promoting MC2R cell surface expression in adrenal cells for corticotropin (ACTH) signaling and potential involvement in adipocyte intracellular trafficking pathways. MRAP forms antiparallel homodimers and heterodimers, engaging with a range of melanocortin receptors.
