NLS-Cas9-EGFP Nuclease
- Description: CRISPR/Cas9 represents an adaptive immune defense system that has evolved over time within bacteria and archaea. The Cas9 nuclease, functioning as an RNA-guided endonuclease, catalyzes the cleavage of double-stranded DNA, making it a potent tool for precise genome editing. The Cas9 protein forms a highly stable ribonucleoprotein (RNP) complex with guide RNA (sgRNA). When equipped with the NLS sequence, the Cas9-RNP complex can promptly localize to the nucleus upon cell entry, eliminating the need for in vivo transcription or translation. Derived from Streptococcus pyogenes, this product is a recombinant SpCas9 Nuclease mutant expressed in Escherichia coli. By incorporating nuclear localization signals (NLS) at both termini of the protein, it is efficiently directed into the nucleus for genome editing purposes. Additionally, an EGFP fluorescent tag is appended after the C’, enabling fluorescence-activated cell sorting (FACS) to enrich for the desired genome edits, thereby significantly reducing the labor and cost associated with single-cell cloning and genotyping in genome editing applications.
