PolymorphprepTM

Polymorphprep™ is a specialized cell separation medium for human polymorphonuclear leukocyte isolation. Unlike conventional density gradient media designed primarily for PBMC isolation, Polymorphprep™ is formulated specifically to enable separation of the denser polymorphonuclear cell fraction from whole blood.
The medium contains sodium diatrizoate and polysaccharide and has a density of 1.113 ± 0.001 g/mL. Its relatively high osmolality promotes water loss and shrinkage of erythrocytes during centrifugation, increasing their effective buoyant density and allowing the erythrocytes to sediment through the medium while PMNs form a distinct cell band.
Polymorphprep™ is particularly useful for research workflows involving human neutrophils, granulocytes, polymorphonuclear leukocytes and peripheral blood cell populations.

Polymorphprep™ uses a density-based separation mechanism that takes advantage of differences in the buoyant densities of blood cell populations.
Human PMNs have a higher buoyant density than mononuclear cells, while erythrocytes have a density that overlaps with the denser leukocyte populations. The elevated osmolality of Polymorphprep™ causes erythrocytes to lose water and shrink during centrifugation, increasing their effective density and promoting their sedimentation through the medium. This allows distinct leukocyte fractions to form during centrifugation.
Following separation, the upper leukocyte band contains mononuclear cells, while the lower leukocyte band contains polymorphonuclear cells.
Compatible Sample
Polymorphprep™ is designed for whole undiluted human blood.
This is an important distinction and should remain visible on the product page. Manufacturer information specifically states that the method is intended for whole undiluted human blood, rather than leukocyte-rich fractions or blood from animal species.
