Target
Transcriptional regulator that controls a genetic switch in male development. It is necessary and sufficient for initiating male sex determination by directing the development of supporting cell precursors (pre-Sertoli cells) as Sertoli rather than granulosa cells (By similarity) . In male adult brain involved in the maintenance of motor functions of dopaminergic neurons (By similarity) . Involved in different aspects of gene regulation including promoter activation or repression (By similarity) . Promotes DNA bending. SRY HMG box recognizes DNA by partial intercalation in the minor groove. Also involved in pre-mRNA splicing. Binds to the DNA consensus sequence 5'-[AT]AACAA[AT]-3'.

Assay Principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Sex Determining Region Y (SRY) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Sex Determining Region Y (SRY) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Sex Determining Region Y (SRY), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Sex Determining Region Y (SRY) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
