Calcium-dependent, calmodulin-stimulated protein phosphatase which plays an essential role in the transduction of intracellular Ca (2+) -mediated signals (PubMed:7791792, PubMed:26794871) . Many of the substrates contain a PxIxIT motif and/or a LxVP motif (By similarity) . In response to increased Ca (2+) levels, dephosphorylates and activates phosphatase SSH1 which results in cofilin dephosphorylation (By similarity) . In response to increased Ca (2+) levels following mitochondrial depolarization, dephosphorylates DNM1L inducing DNM1L translocation to the mitochondrion (By similarity) . Dephosphorylates heat shock protein HSPB1 (By similarity) . Dephosphorylates and activates transcription factor NFATC1 (By similarity) . Assay Principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Calcineurin (CaN) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Calcineurin (CaN) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Calcineurin (CaN), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Calcineurin (CaN) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
