Target
Required for TCR (T-cell antigen receptor) - and pre-TCR-mediated signaling, both in mature T-cells and during their development. Involved in FCGR3 (low affinity immunoglobulin gamma Fc region receptor III) -mediated signaling in natural killer cells and FCER1 (high affinity immunoglobulin epsilon receptor) -mediated signaling in mast cells. Couples activation of these receptors and their associated kinases with distal intracellular events such as mobilization of intracellular calcium stores, PKC activation, MAPK activation or cytoskeletal reorganization through the recruitment of PLCG1, GRB2, GRAP2, and other signaling molecules.

Assay Principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Linker For Activation Of T-Cell (LAT) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Linker For Activation Of T-Cell (LAT) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Linker For Activation Of T-Cell (LAT), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Linker For Activation Of T-Cell (LAT) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
