CDK8 ELISA Kit (Human)

  • Reproducibility :

Intra-assay Precision (Precision within an assay) : 3 samples with low, middle and high level Cyclin Dependent Kinase 8 (CDK8) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays) : 3 samples with low, middle and high level Cyclin Dependent Kinase 8 (CDK8) were tested on 3 different plates, 8 replicates in each plate. CV (%) = SD/meanX100 Intra-Assay: CV<10% Inter-Assay: CV<12%
  • Gene Name :

    Cyclin dependent kinase 8
  • Gene Aliases :

CDK8 protein kinase; cell division protein kinase 8; cyclin-dependent kinase 8; IDDHBA; K35; mediator complex subunit CDK8; mediator of RNA polymerase II transcription subunit CDK8; protein kinase K35.
  • Gene ID :

    1024
  • Accession Number :

    NP_001251.1
  • Reactivity :

    Homo sapiens|Human
  • Target :

Component of the Mediator complex, a coactivator involved in regulated gene transcription of nearly all RNA polymerase II-dependent genes. Mediator functions as a bridge to convey information from gene-specific regulatory proteins to the basal RNA polymerase II transcription machinery. Mediator is recruited to promoters by direct interactions with regulatory proteins and serves as a scaffold for the assembly of a functional preinitiation complex with RNA polymerase II and the general transcription factors. Phosphorylates the CTD (C-terminal domain) of the large subunit of RNA polymerase II (RNAp II), which may inhibit the formation of a transcription initiation complex. Phosphorylates CCNH leading to down-regulation of the TFIIH complex and transcriptional repression. Recruited through interaction with MAML1 to hyperphosphorylate the intracellular domain of NOTCH, leading to its degradation.
  • Type :

    ELISA Kit
  • Applications :

    Enzyme-linked Immunosorbent assay-Sandwich
  • Detection Method :

    Colorimetric
  • Assay Principle :

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Cyclin Dependent Kinase 8 (CDK8) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Cyclin Dependent Kinase 8 (CDK8) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Cyclin Dependent Kinase 8 (CDK8), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Cyclin Dependent Kinase 8 (CDK8) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
