Target
Proteinase 3 also known as PRTN3 is an enzyme that in humans is encoded by the PRTN3 gene. This gene is mapped to 19p13.3. PRTN3 is a serine protease enzyme expressed mainly in neutrophil granulocytes. Its exact role in the function of the neutrophil is unknown, but, in human neutrophils, proteinase 3 contributes to the proteolytic generation of antimicrobial peptides. It is also the target of anti-neutrophil cytoplasmic antibodies (ANCAs) of the c-ANCA (cytoplasmic subtype) class, a type of antibody frequently found in the disease granulomatosis with polyangiitis.

Assay Principle
Aviva's human Myeloblastin ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Myeloblastin has been precoated onto 96-well plates. Standards (Expression system for standard: E.coli, Immunogen sequence: I28-R248) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Myeloblastin is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The absorbance of yellow is proportional to the human Myeloblastin amount of sample captured in plate.
