  • Tag:

    N-terminal 6xHis-SUMO-tagged
  • Type:

    Developed Protein
  • Source:

    E.coli
  • Field of Research:

    Signal Transduction
  • Relevance:

Involved in several stages of intracellular trafficking. Interacts with mbranes containing phosphatidylinositol 3-phosphate (PtdIns (3P) ) or phosphatidylinositol 3,5-bisphosphate (PtdIns (3,5) P2) . Acts in part as component of the retromer mbrane-deforming SNX-BAR subcomplex. The SNX-BAR retromer mediates retrograde transport of cargo proteins from endosomes to the trans-Golgi network (TGN) and is involved in endosome-to-plasma mbrane transport for cargo protein recycling. The SNX-BAR subcomplex functions to deform the donor mbrane into a tubular profile called endosome-to-TGN transport carrier (ETC) (Probable) . Can sense mbrane curvature and has in vitro vesicle-to-mbrane rodeling activity . Involved in retrograde endosome-to-TGN transport of lysosomal enzyme receptors (IGF2R, M6PR and SORT1) and Shiginella dysenteria toxin stxB. Plays a role in targeting ligand-activated EGFR to the lysosomes for degradation after endocytosis from the cell surface and release from the Golgi . Involvent in retromer-independent endocytic trafficking of P2RY1 and lysosomal degradation of protease-activated receptor-1/F2R . Promotes KALRN- and RHOG-dependent but retromer-independent mbrane rodeling such as lamellipodium formation; the function is dependent on GEF activity of KALRN . Required for endocytosis of DRD5 upon agonist stimulation but not for basal receptor trafficking .
  • Endotoxin:

    Not test
  • Purity:

Greater than 90% as determined by SDS-PAGE.
  • Activity:

    Not Test
  • Form:

    Liquid or Lyophilized powder
  • Buffer:

If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
  • Reconstitution:

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
  • Function:

Involved in several stages of intracellular trafficking. Interacts with membranes containing phosphatidylinositol 3-phosphate (PtdIns (3P) ) or phosphatidylinositol 3,5-bisphosphate (PtdIns (3,5) P2)

  • Storage Conditions:

The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself. Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
