Storage_stabilityThe purified (UNLB) avidin is supplied as 5.0 mg protein in 1.0 mL of 100 mM borate buffered saline, pH 8.2. No preservatives or amine-containing buffer salts added. Store at 2-8 degree C. The fluorescein (FITC), rhodamine (TRITC), Texas Red (TXRD), Cyanine 2 (CY 2), and Cyanine 5 (CY 5) conjugates are supplied as 1.0 mg in 1.0 mL PBS/NaN3. Store at 2-8 degree C. The Cyanine 3 (CY 3) conjugate is supplied as 0.1 mg in 0.2 mL PBS/NaN3. Store at 2-8 degree C. The alkaline phosphatase (AP) conjugate is supplied as 1.0 mL of stock solution in 50mM Tris/1mM MgCl2/50% Glycerol, pH 8.0, containing 0.1% NaN3 as preservative. Store at 2-8 degree C or long-term at -20 degree C. The horseradish peroxidase (HRP) conjugate is supplied as 1.0 mL of stock solution in 50% glycerol/50% PBS, pH 7.4. No preservative added. Store at 2-8 degree C or long-term at -20 degree C. The beta -galactosidase (BGAL) conjugate is supplied as 1.0 mL of stock solution in 50% glycerol/50% PBS, pH 7.4, containing 0.1% sodium azide as preservative. Store at 2-8 degree C or long-term at -20 degree C. The R-phycoerythrin (R-PE) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze! The allophycocyanin (APC) conjugate is supplied as 0.1 mg in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze! The Spectral Red (SPRD) conjugate is supplied as 0.1 mg in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze! Protect conjugated forms from light. PropertiesThis MyBioSource Fluorescein isothiocyanate (FITC) antibody is currently after some BD antibodies the most commonly used fluorescent dye for FACS. When excited at 488 nanometers, FITC has a green emission that's usually collected at 530 nanometers, the FL1 detector of a FACSCalibur or FACScan. FITC has a high quantum yield (efficiency of energy transfer from absorption to emission fluorescence) and approximately half of the absorbed photons are emitted as fluorescent light. For fluorescent microscopy applications, the 1 FITC is seldom used as it photo bleaches rather quickly though in flow cytometry applications, its photo bleaching effects are not observed due to a very brief interaction at the laser intercept. MyBioSource FITC is highly sensitive to pH extremes.

ConceptScope note: Test for tissue antigen using either a direct method, by conjugation of antibody with fluorescent dye (FLUORESCENT ANTIBODY TECHNIQUE, DIRECT) or an indirect method, by formation of antigen-antibody complex which is then labeled with fluorescein-conjugated anti-immunoglobulin antibody (FLUORESCENT ANTIBODY TECHNIQUE, INDIRECT). The tissue is then examined by fluorescence microscopy.
