Long description
The reactivity of the antiserum is restricted to haptoglobin. In immunoelectrophoresis and radial immunodiffusion (Ouchterlony), using various antiserum concentrations against fresh normal swine plasma a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified haptoglobin. No reaction is obtained with any other plasma protein component or serum. In precipitating techniques as immunoelectrophoresis and single or double radial immunodiffusion to identify the presence of haptoglobin in swine plasma or other body fluids or to determine its concentration. The presence of non-precipitating antibodies has not been assayed. This does not exclude the use in non-precipitating antibody-binding techniques if proper controls are included.

Antibody come from
Haptoglobin is an acute phase protein synthesized in the liver. In immunoelectrophoresis it has an alpha-2 mobility. It forms a group of heterogeneous proteins consisting of four peptide chains, two alpha and two beta chains. The molecular weight is about 86,000. Haptoglobin combines with haemoglobin and is then rapidly catabolized. Concentration is elevated in cases of inflammation and reduced in liver disease and haemolytic anaemia. Freund’s complete adjuvant is used in the first step of the immunization procedure.

Other description
Delipidated, heat inactivated, lyophilized, stable whole antiserum No preservative added Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal goat serum. No foreign proteins added.

Antibody s reacts with these species
The antiSerum does not cross-react with any other component of Swine plasma. Inter-species crossreactivity is a normal feature of antibodies to plasma proteins since they frequently share antigenic determinants. Cross-reactivity of this antiSerum has been tested in double radial immunodiffusion anti several sera with the following results: Bovine - Cat - Chicken - Dog - Duck - Guinea Pig - Hamster - Horse - Human - Monkey - Mouse - pigeon - Rat - Sheep - Turkey-

Antibody s suited for
Immunoprecipitation. In immunoelectrophoresis use 2 ml or equivalent against 120 ml antiserum. In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 ml in a 3 mm diameter center well and 2 ml serum samples (neat and serially diluted) in 2 mm diameter peripheral wells. Precipitin titre 1:32 when tested against pooled normal swine plasma in agar-block immunodiffusion titration.

Storage
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. Reconstitute the lyophilized antiserum by add ing 1 ml sterile distilled water. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4°C, not ref rozen, and preferably used the same day. Lyophilized at +4°C--at least 10 years. Reconstituted at or below -20°C--3-5 years. Reconstituted at +4°C--7 days

Description
This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided.

Host
Goat, The goat polyclonal antibodies are directed against human genes or rabbit or mouse IgGs (H +L) chain primary antibodies and conjugated for detection. It are very good secondary antibodies that are affinity purified and have a good affinity themselves too. Often KLH is coupled to the antigen so anti KLH can be present in the vial.
