Flow cytometry uses monoclonal antibodies of specific affinity clones for cell counting, cell sorting and biomarker detection by suspending cells in a stream of fluid for Forward Scatter, FSC and side scatter, SSC analysis. Human PBMCs can be loaded with CFSE tracking dye after non adherent cell harvesting. Subsequently labeled with anti-CD antibodies, and analyzed by multiparameter flow cytometry. Two-parameter profiles of CD vs. CFSE; and another CD vs. FSC-W. We suggest to use FSC-H vs. FSC-A. FSC-A, FSC-H, FSC-W = area, height, and width of the forward 488 nm light scatter from the flow signal.

Interleukin 8 (IL8 or chemokine (C-X-C motif) ligand 8, CXCL8) is a chemokine produced by macrophages and other cell types such as epithelial cells, airway smooth muscle cells and endothelial cells. Endothelial cells store IL-8 in their storage vesicles, the Weibel-Palade bodies. In humans, the interleukin-8 proteins encoded by the IL8 gene. IL-8 is initially produced as a precursor peptide of 99 amino acids long which then undergoes cleavage to create several active IL-8 isoforms. In culture, a 72 amino acid peptide is the major form secreted by macrophages. Recombinant, rec. E. coli interleukin-8 for cell culture or antibody production.

Induced protein genes, factors or kinases, increase the production of (an enzyme or other protein) at the level of genetic transcription. The receptors are ligand binding factors of type 1, 2 or 3 and protein-molecules that receive chemical-signals from outside a cell. When such chemical-signals couple or bind to a receptor, they cause some form of cellular/tissue-response, e.g. a change in the electrical-activity of a cell. In this sense, am olfactory receptor is a protein-molecule that recognizes and responds to endogenous-chemical signals, chemokinesor cytokines e.g. an acetylcholine-receptor recognizes and responds to its endogenous-ligand, acetylcholine. However, sometimes in pharmacology, the term is also used to include other proteins that are drug-targets, such as enzymes, transporters and ion-channels.

Scope note:
In vitro method for producing large amounts of specific DNA or RNA fragments of defined length and sequence from small amounts of short oligonucleotide flanking sequences (primers). The essential steps include thermal denaturation of the double-stranded target molecules, annealing of the primers to their complementary sequences, and extension of the annealed primers by enzymatic synthesis with DNA polymerase. The reaction is efficient, specific, and extremely sensitive. Uses for the reaction include disease diagnosis, detection of difficult-to-isolate pathogens, mutation analysis, genetic testing, DNA sequencing, and analyzing evolutionary relationships.
