ELISA kit description
GENTAUR's rat NGF ELISA test is a ready to use ELISA kit. We use a monoclonal from mouse specific for NGF was used as capure antibodies on the ELISA test plate. The internal standards (NSO, S122-G241) and your samples are measured with a biotin conjugated polyclonal from goat specific for NGF will than be washed with PBS. The colour reaction with TMB on HRP will colour the well from blue to yellow with the stop solution. The more rat NGF in your sample the higher the ELISA signal will be compared to this elisa's standard curve.

How the elisa works
Sample the amount of 100ul/well of the standard solution at 1000 pg per ml, 500, 25, 125, 62.5, 31.25, 15.6 pg/ml rat NGF standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffers into the control well (Zero well). Add 0.1ml of each properly diluted sample of rat cell culture supernatants or serum to the ELISA plate. Keep in mind that each rat NGF standard solution and each sample be measured in duplicate.

Nerve growth factor(NGF) is a polypeptide involved in the regulation of growth and differentiation of sympathetic and certain sensory neurons. NGF is thought to have a profound effect on the development and maintenance of sympathetic and embryonic sensory neurones. NGF activity isolated from the male mouse submaxillary gland(MSG) consists of three types of subunits, alpha, beta and gamma, which specifically interact to form a 7S, approximately 130,000-molecular weight(Mr) complex. The 7S complex contains two identical 118-amino acid beta-chains, which are solely responsible for the nerve growth-stimulating activity of NGF. NGF, which is expressed by inflammatory cells and effects changes that lead to increased neural responsiveness, could be a pivotal mediator in allergic rhinitis.
