ELISA kit description
GENTAUR's mouse CXCL13 ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibodies from rat specific for CXCL13 was used as capure antibodies on the ELISA test plate. The internal standards (E.coli, I22-A109) and your samples are measured with a biotin conjugated polyclonal from goat specific for CXCL13 is added subsequently and then followed by washing with PBS or TBS buffers. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffers. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse CXCL13 in your sample the higher the ELISA signal will be compared to this elisa's standard curve.

How the elisa works
Sample the amount of 100ul/well of the standard solution at 1000 pg per ml, 500, 25, 125, 62.5, 31.25, 15.6 pg/ml mouse CXCL13 standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffers into the control well (Zero well). Add 0.1ml of each properly diluted sample of mouse cell culture supernatant, serum or plasma(heparin, EDTA) to the ELISA plate. Keep in mind that each mouse CXCL13 standard solution and each sample be measured in duplicate.

C-X-C motif chemokine 13(CXCL13), also known as B lymphocyte chemoattractant(BLC), is a protein that in humans is encoded by theCXCL13 gene. CXCL13 is a small cytokine belonging to the CXC chemokine family, and it was located on sequence maps to 4q21, near the genes encoding most CXC chemokines. In T-lymphocytes, CXCL13 expression is thought to reflect a germinal center origin of the T-cell. Hence, expression of CXCL13 in T-cell lymphomas, such as Angioimmunoblastic T-cell Lymphoma, is thought to reflect a germinal center origin of the neoplastic T-cells.
