ELISA kit description
GENTAUR's human Legumain(total) ELISA test is a ready to use ELISA kit. We use a monoclonal from mouse specific for Legumain(total) was used as capure antibodies on the ELISA test plate. The internal standards (Expression system for standard: NSO, Immunogen sequence: V18-Y433) and your samples are measured with a biotin conjugated polyclonal from goat specific for Legumain(total) will than be washed with PBS. The colour reaction with TMB on HRP will colour the well from blue to yellow with the stop solution. The more human Legumain(total) in your sample the higher the ELISA signal will be compared to this elisa's standard curve.

How the elisa works
Sample the amount of 100ul/well of the standard solution at 2000pg/ml, 1000pg/ml, 500pg/ml, 250pg/ml, 125pg/ml, 62.5pg/ml, 31.2pg/ml human Legumain(total) standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffers into the control well (Zero well). Add 0.1ml of each properly diluted sample of human cell culture supernatant, cell lysate, tissue homogenates, serum or plasma (heparin, EDTA) to the ELISA plate. Keep in mind that each human Legumain(total) standard solution and each sample be measured in duplicate.

Legumain (asparaginyl endopeptidase, citvac, proteinase B, hemoglobinase, PRSC1 gene product or LGMN (Homo sapiens), vicilin peptidohydrolase, bean endopeptidase) is an enzyme that in humans is encoded by the LGMN gene (previous symbolPRSC1). Using fluorescence in situ hybridization, the LGMN gene was mapped to chromosome 14q32.1. This enzyme may be involved in the processing of bacterial peptides and endogenous proteins for MHC class II presentation in the lysosomal/endosomal systems. Enzyme activation is triggered by acidic pH and appears to be autocatalytic. Protein expression occurs after monocytes differentiate into dendritic cells. A fully mature, active enzyme is produced following lipopolysaccharide expression in mature dendritic cells. Overexpression of this gene may be associated with the majority of solid tumor types.
