ELISA kit description
GENTAUR's human CCL19 ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibodies from mouse specific for CCL19 was used as capure antibodies on the ELISA test plate. The internal standards (E.coli, G22-S98) and your samples are measured with a biotin conjugated polyclonal from goat specific for CCL19 is added subsequently and then followed by washing with PBS or TBS buffers. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffers. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human CCL19 in your sample the higher the ELISA signal will be compared to this elisa's standard curve.

How the elisa works
Sample the amount of 100ul/well of the standard solution at 1000 pg per ml, 500, 25, 125, 62.5, 31.25, 15.6 pg/ml human CCL19 standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffers into the control well (Zero well). Add 0.1ml of each properly diluted sample of human cell culture supernatant, serum or plasma(heparin, EDTA) to the ELISA plate. Keep in mind that each human CCL19 standard solution and each sample be measured in duplicate.

Chemokine(C-C motif) ligand 19(CCL19) is a small cytokine belonging to the CC chemokine family that is also known as EBI1 ligand chemokine(ELC) and macrophage inflammatory protein-3-beta (MIP-3-beta). Using PCR of human-rodent hybrids, the CCL19 gene was map to human chromosome 9p13, and the CCL19 gene is not part of the CC chemokine gene cluster on chromosome 17. The cytokine encoded by this gene may play a role in normal lymphocyte recirculation and homing. It also plays an important role in trafficking of T cells in thymus, and in T cell and B cell migration to secondary lymphoid organs.
