Murine CD30, Mr 52 kDa (core peptide) CD30 is a type I transmembrane protein and a member of the tumor necrosis factor receptor superfamily of proteins.^1,2 Murine CD30 is expressed predominantly in the thymus, and it is inducible in mouse splenocytes stimulated with pokeweed mitogen or concanavalin A.^1 In anti-CD3 epsilon -activated spleen cells, CD30 is expressed primarily on the surface of CD8^+ T cells with peak expression on days 4 and 5. Stimulation of CD30^+ CTL lines with plate-bound anti-CD30 directly signals IL-5 but not IFN- gamma production. ^1 While these studies demonstrate that CD30 directs cytokine secretion and suggest that CD30 may play a pivotal role in the pattern of cytokine production by T cells, the precise roles of CD30 and its ligand (CD153) in T-cell development have not been clearly defined.

The purified (UNLB) antibody is supplied as 0.5 mg of purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.2. No preservatives or amine-containing buffer salts added. Store at 2- 8 degree C. The fluorescein (FITC) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C. The biotin (BIOT) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C. The R-phycoerythrin (R-PE) conjugate is supplied as 0.1 mg in 1.0 mL or 0.2 mg in 2.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze! The low endotoxin, azide-free (LE/AF) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of PBS. Aliquot and store at or below -20 degree C. Protect conjugated forms from light. Aliquot and freeze the low endotoxin, azide-free product at -20 degree C immediately upon receipt. Each reagent is stable for the period shown on the bottle label if stored as directed.

This MyBioSource Fluorescein isothiocyanate (FITC) antibody is currently after some BD antibodies the most commonly used fluorescent dye for FACS. When excited at 488 nanometers, FITC has a green emission that's usually collected at 530 nanometers, the FL1 detector of a FACSCalibur or FACScan. FITC has a high quantum yield (efficiency of energy transfer from absorption to emission fluorescence) and approximately half of the absorbed photons are emitted as fluorescent light. For fluorescent microscopy applications, the 1 FITC is seldom used as it photo bleaches rather quickly though in flow cytometry applications, its photo bleaching effects are not observed due to a very brief interaction at the laser intercept. MyBioSource FITC is highly sensitive to pH extremes. Armenian of Syrian hamster monoclonal IgGs are useful antibodies for double staining your IHC samples. Using mabs or monoclonal antibodies has a lot of advantages for Flow cytometry.

The tissue is then examined by fluorescence microscopy.
