Kronos HT

High throuput assay system for real-time gene expression monitoring of living cells.

WSL-1565 Kronos HT is the high throughput support model of the assay system that measure the gene expression of living cells in real-time with the bioluminescence of luciferase. Kronos Dio, which is a precedent model compatible with 35 mm dish, has been used as a real-time luminescence measurement system with cell culturing function, but this time a new lineup of model compatible with multiwell plate measurement is added. With this new product Kronos HT, we succeeded the know-how cultivated in the precedent model in order to measure the luminescence while culturing the cells, and further strengthened functions and performance for high throughput measurement. Please consider adopting Kronos HT for high-throughput measurement in biological clock analysis, change in drug response with time, cytotoxicity test etc.

Reporter assay in real time with culturing cells

Reporter assay is used to measure the transcriptional activity of gene transcription regulatory sequences such as promoters, for researches on drug response and gene expression control, evaluation of gene transfer efficiency, and so on. Among them, the method of measuring the amount of bioluminescence using bioluminescent enzyme luciferase using a device called a luminometer is superior in terms of high sensitivity and low background and is widely used. Usually, transcription activity at a certain point (end point) is measured, but the assay is also becoming used for tracking transcription activity over time such as stimulation response, time course of transgene expression level, periodic variation of biological clock gene. Kronos HT is a system to optimize for such assays with the ability to monitor the bioluminescence over time as cells are cultured.

The “WSP-3310 peristaQuantumPump” operates within a wide range of 0.01 rpm to 50 rpm (a maximum adjustment range of 5,000 times), where the flow rate is proportional to the pump’s rotation speed, exhibiting consistent linearity (R-squared coefficient = 1). In long-term liquid delivery experiments conducted at set flow rates from 1 mL/min to 40 mL/min, excluding the initial 1 hour until the tube stabilized, it was confirmed that very stable flow control is possible with a CV value of 1.07%. Additionally, upon evaluating the effects of liquid temperature, it was demonstrated that stable liquid delivery can be achieved under consistent temperatures. These results indicate that the peristaQuantumPump can achieve high precision and stable liquid delivery across a broad range of flow rates.

 

Innovative technology

Fluid control with PeristaQuantumPump

 

PeristaltQuantumPump, likes other tube-based pumps, utilize the elasticity of the tube to transport fluids.

The core mechanism of this pump involves the continuous compression of the tube by rollers moving along it. This action, combined with the pressure exerted and the tube’s restorative force, propels the fluid. PeristaltQuantumPump employs self-propelled rollers. When the main shaft (A) rotates, the surrounding rollers (B) move in the direction of the shaft’s rotation. Upon contacting the tube, rollers (B) rotate in the opposite direction of the main shaft (A) while compressing the tube. As the tube passes through the compression section, a negative pressure is created inside, drawing in liquid, and the rotational motion expels the liquid on the outlet side. Since the liquid only travels inside the tube and does not come into contact with other parts of the pump, the risk of contamination is extremely low. Additionally, simply replacing the tube allows for easy cleaning and maintenance. The elasticity of the tube also enables the transport of viscous liquids or liquids containing particles. PeristaltQuantumPump offers precise flow control and high reproducibility, making them suitable for a wide range of applications in the medical, research, and industrial fields.

This is the original peristaQuantumPump, a condensation of Ato’s technological essence. With 12 rollers minimizing the effects of pulsation, it achieves reproducible liquid delivery with high flow rate precision. Proper compression control extends the tube’s lifespan to over two weeks, even with continuous operation at maximum flow rate, while maintaining flow rate precision and stability. Utilizing three flow paths, it allows for the delivery of multiple targets under identical conditions with even flow rates or the delivery of multiple solutions at the same flow rate. Moreover, with the PQP support program, external control can be easily programmed. WSE-5300 Printgraph CMOSI is High resolution 6M pixels monochrome CMOS camera applied with touch panel control system. Touch panel interface is reflecting ATTO original knowhow which is closing user-friendly for 30 years sales. Detecting by UV light source which DNA(EtBr) and detection CBB pattern with white flat LED light source. ATTO original「CyanRed Epi」is switchable LED light which are Cyan LED and Red LED. Cyan LED is available to detect a fluorescence in both blue and green wavelenth. Detetable EtBr which is difficult to detect in Blue LED. However, ATTO made its possible for UV free Gel Documentation System with CyanRed Epi. Save an image to USB memory directly or print out immediately with UP-X898MD thermal printer.

Normalization of Western Blotting Using Total Protein

🔍 Why Normalize Western Blots?

Western blotting involves many variables. Even if the same amount of sample is applied to each lane, differences in sample type, pipetting accuracy, antibody reaction, or transfer method can affect the results. Normalization corrects these inconsistencies, allowing reliable comparison between samples or blots.

⚠️ Limitations of Housekeeping Proteins

Traditionally, HKPs like GAPDH, actin, or tubulin are used as internal controls. However, their expression levels can vary between tissue types, developmental stages, or under different experimental conditions. Moreover, their high abundance can mask the true expression level of the target protein, leading to incorrect conclusions.

Advantages of Total Protein Normalization (TPN)

Using total protein as a reference offers a more stable and universal approach. It reflects the actual amount of protein transferred per lane, reducing the risk of error caused by variation in individual protein expression or antibody reactivity. TPN is compatible with modern labeling techniques, such as fluorescent dyes and stain-free gels.

🔧 How to Perform TPN

Start by labeling protein samples using fluorescent reagents like EzLabel FluoroNeo, or use stain-free gels. Avoid adding reducing agents before labeling, as they interfere with the reaction.

Run the samples on SDS-PAGE and transfer the proteins onto a PVDF membrane. Fluorescence-compatible, low-background membranes are preferred. After transfer, image the membrane to detect total protein, and then proceed with blocking and antibody-based detection of the target protein.

Use software such as CS Analyzer 4 to analyze the images. Define each lane and band, subtract the background, and then calculate the normalized intensity of the target protein based on the corresponding total protein signal in the same lane.

💻 Normalization Method

To calculate the normalized value of a target protein, divide the band intensity of the target protein by the total protein reference value for that lane. This value is then compared against a reference lane set as “1” to determine relative expression levels.

📈 Interpreting Results

Normalization using total protein usually shows consistent and expected trends, such as clear dilution patterns or accurate changes in expression. In contrast, HKP-based normalization can sometimes give misleading results, especially if HKPs are overexpressed or affected by experimental conditions.

🔬 Tools & Reagents Used

The method commonly uses reagents like EzLabel FluoroNeo for protein labeling and EzApply for sample preparation. Transfer buffers like EzFastBlot ensure fast and efficient protein transfer. PVDF membranes with low fluorescence background are ideal for imaging. Stain-free gels or dyes like Ponceau and CBB can also be used for total protein detection.

🔄 Alternative Detection Approaches

Other than fluorescent labeling, stain-free gels can also visualize total protein through UV activation. Membranes stained with CBB or Ponceau provide visible detection, although they may have lower sensitivity compared to fluorescent methods.
