Specificity
Keratin 18 (48 kDa)1 Keratins are a family of intermediate filament proteins that assemble into filaments through forming heterodimers of one type I keratin (keratins 9 to 23) and one type II keratin (keratins 1-8). The two keratin types share only 30% sequence homology.2-6 Keratins demonstrate tissue- and differentiation-specific expression profiles. Cytokeratin 18, also known as keratin 18, is a type I intermediate filament protein of approximately 48 kDa. It exists as a heterotetramer composed of two type I and two type II keratins. Cytokeratin 18 is expressed in all simple type epithelia and basal cells of many squamous and non-epidermal epithelia, and is often co-expressed with cytokeratin 8.7-9 Tissues from the gastrointestinal tract, respiratory tract and urogenital tract, as well as endocrine and exocrine tissues and mesothelial cells are positive for cytokeratin 18. The SB38b monoclonal antibody reacts specifically with keratin 18 and recombinant N-terminal keratin 18.

Storage_stability
The purified (UNLB) antibody is supplied as 0.1 mg purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.2; store at 2-8 degree C. The fluorescein (FITC) conjugate is supplied as 0.1 mg in 0.2 mL PBS/NaN3. Store at 2-8 degree C. The horseradish peroxidase (HRP) conjugate is supplied as 1.0 mL of stock solution in 50% glycerol/50% PBS, pH 7.4. No preservative added. Store at 2-8 degree C or long-term at -20 The biotin (BIOT) conjugate is supplied as 0.1 mg in 0.2 mL PBS/NaN3. Store at 2-8 degree C.

Description
This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided. Antibody for research use.

Properties
This MyBioSource Fluorescein isothiocyanate (FITC) antibody is currently after some BD antibodies the most commonly used fluorescent dye for FACS. When excited at 488 nanometers, FITC has a green emission that's usually collected at 530 nanometers, the FL1 detector of a FACSCalibur or FACScan. FITC has a high quantum yield (efficiency of energy transfer from absorption to emission fluorescence) and approximately half of the absorbed photons are emitted as fluorescent light. For fluorescent microscopy applications, the 1 FITC is seldom used as it photo bleaches rather quickly though in flow cytometry applications, its photo bleaching effects are not observed due to a very brief interaction at the laser intercept. MyBioSource FITC is highly sensitive to pH extremes.

Test
Mouse or mice from the Mus musculus species are used for production of mouse monoclonal antibodies or mabs and as research model for humans in your lab. Mouse are mature after 40 days for females and 55 days for males. The female mice are pregnant only 20 days and can give birth to 10 litters of 6-8 mice a year. Transgenic, knock-out, congenic and inbread strains are known for C57BL/6, A/J, BALB/c, SCID while the CD-1 is outbred as strain.

Concept
Scope note:
Test for tissue antigen using either a direct method, by conjugation of antibody with fluorescent dye (FLUORESCENT ANTIBODY TECHNIQUE, DIRECT) or an indirect method, by formation of antigen-antibody complex which is then labeled with fluorescein-conjugated anti-immunoglobulin antibody (FLUORESCENT ANTIBODY TECHNIQUE, INDIRECT). The tissue is then examined by fluorescence microscopy.
