Background:
The reactivity of the antiserum is restricted to alpha-1 antichymotrypsin. In immunoelectrophoresis and radial immunodiffusion, using various antiserum concentrations against Human plasma a single precipitin line is obtained and shows a reaction of full identity with the purified immunogen. In precipitating techniques as immunoelectrophoresis and single and double radial immunodiffusion (Mancini, Ouchterlony) to identify the presence of alpha-1 antichymotrypsin or to determine its concentration. The presence of non-precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in non-precipitating antibody-binding techniques if proper controls are included.UniProt:

Alpha-1 antichymotrypsin is a protease inhibitor with a molecular weight of 68.000. Its concentration in fetal Human serum is low at birth, but the concentration increases rapidly to normal plasma levels of about is 4.5 μg/mL. Alpha-1 antichymotrypsin is regulated like a an acute phase reactant. It has been isolated as a homogenous protein for use in the antiserum production. Freund’s complete adjuvant is used in the first step of the immunization procedure.Assay Principle:
Precipitation assays. In immunoelectrophoresis in agarose-plates use 2 μl Human plasma or equivalent against 120 μl antiserum. In double radial immunodiffusion use a rosette arrangement with 10 μl antiserum in 3 mm diameter center well and 2 μl plasma samples (neat and serially diluted) in 2 mm diameter peripheral wells.Form:
