Description
RPMI-1640 With L-Glutamine, 25mM HEPES buffer and 2.2gms per litre Sodium bicarbonate, is an orangish red colored, clear solution. RPMI were developed for the culture of human normal and neoplastic cells in vitro. It was specifically designed to support the growth of human lymphoblastoid cells in suspension culture. Now the medium is extensively used for a wide range of anchorage dependant cell lines. It is also known to support growth of cells in the absence of serum.

Cultural Response

Buffering solutions are useful to keep the pH range sable when using this reagent of Himedia. The Liter or liter is a metric system unit of volume equal to 1 cubic decimeter (dm3), 1,000 cubic centimeters (cm3) or 1/1,000 cubic meter. A cubic decimeter (or liter) occupies a volume of 10×10×10 centimeters (see figure) and is thus equal to one-thousandth of a cubic meter. Water baths of various volumes are available for lab research.

Buffer
25mM HEPES (4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid ) pH7 solution buffers are a media with a zwitterion organic chemical buffering agent; one of the twenty Good's buffers. HEPES DMEM medium is widely used in cell culture, largely because it is better at maintaining physiological pH 7 tot 7,5 despite changes in carbon dioxide concentration (produced by cellular respiration) when compared to bicarbonate buffers, which are also commonly used in cell culture. The dissociation of water decreases with falling temperature, but the dissociation constants (pK) of many other buffers do not change much with temperature. HEPES, 25 mM in RPMI and DMEM is like water in that its dissociation decreases as the temperature decreases. This makes HEPES RPMI 25 milli-molar media with or without L-glutamine a more effective buffering agent for maintaining enzyme structure and function at low temperatures.
