Scientific context
The Tumor Necrosis Factor Receptor (TNFR) also known as Cluster of differentiation (CD120) is a protein that belongs to the (TNF)/ (TNFR) superfamily. TNF interacts with two distinct receptors TNFR1 and TNFR2. These receptors share no homology on their cytoplasmic sequences(1,3).TNFR1 also known as p55/p60 is a high affinity receptor for TNF-α. The TNFR1 has an extracellular domain with variable numbers of cysteine-rich repeats. The functional properties of TNFR1 are targets in new therapies for osteoporosis, chronic inflammatory and autoimmune diseases (1, 2). The TNF-α/TNFR1 receptor complex is responsible for the recruitment and the subsequent activation of the caspase (aspartate-specific cysteine proteases) that regulate apoptosis.

Test
Mouse or mice from the Mus musculus species are used for production of mouse monoclonal antibodies or mabs and as research model for humans in your lab. Mouse are mature after 40 days for females and 55 days for males. The female mice are pregnant only 20 days and can give birth to 10 litters of 6-8 mice a year. Transgenic, knock-out, congenic and inbread strains are known for C57BL/6, A/J, BALB/c, SCID while the CD-1 is outbred as strain.

About
Polyclonals can be used for Western blot, immunohistochemistry on frozen slices or parrafin fixed tissues. The advantage is that there are more epitopes available in a polyclonal antiserum to detect the proteins than in monoclonal sera. Rabbits are used for polyclonal antibody production by StressMark antibodies. Rabbit antibodies are very stable and can be stored for several days at room temperature. StressMark antibodies adds sodium azide and glycerol to enhance the stability of the rabbit polyclonal antibodies. Anti-human, anti mouse antibodies to highly immunogenic selected peptide sequences are" monoclonal like" since the epitope to which they are directed is less than 35 amino acids long.

Gene
Tumor necrosis factor (TNFa, tumor necrosis factor alpha, TNFα, cachexin, or cachectin) is a cell signaling protein (cytokine) involved in systemic inflammation and is one of the cytokines that make up the acute phase reaction. It is produced chiefly by activated macrophages, although it can be produced by many other cell types such as CD4+ lymphocytes, NK cells, neutrophils, mast cells, eosinophils, and neurons. TNFb or TNF beta also bin on TNF receptors for Th1 activation.

Technique
Polyclonal, Rabbit, anti-, Mouse, anti, antibody to, antibodies, mouses, Polyclonal antibodies (pAbs) are mostly rabbit or goat antibodies that are secreted by different B cells, whereas monoclonal antibodies come from a single N cell lineage. Pabs are a collection of immunoglobulin molecules that react against a specific antigen, each identifying a different epitope.
